Multi Target Cell Separation - pluribead Cascade

Simultaneous MTS is an easy and fast way to isolate more than one target from a cell suspension at the same time. To this end, the beads that bind to various cell types are differently sized.

Within the cascade mode, sieves are piled so that the top sieve holds back the bigger beads, while the lower sieve holds back the smaller beads with the respective target cells. Thus, for example, CD4 and CD8 cells can be separated from sample material simultaneously.

Three steps characterize the sorting procedure:

  1. Mix pluriBeads and sample material
  2. Pour mixture onto piled sieves, wash off unbound and unwanted cells
  3. Split the cascade and recover your single cell suspension

A special sample preparation is not required.

Sample and beads with different size and specificity are mixed and incubated at room temperature on a roller mixer or pluriPlix. The target cells will bind to the beads. The target cells – now bound to the beads – are separated with pluriSelect‘s cascade technology. With target cells rosetted beads remain onto the sieve. The upper sieve holds back the big pluriBeads, while the lower sieve holds back the small pluriBeads. Washing removes all unspecific contamination. Split the sieves. Detach cells from the beads with a detachment buffer. The cells will be released from the beads directly onto the sieve. Bead free cells will run into tube, beads remain onto the sieve.
