Leukocytes, the white blood cells, are a central component of the immune system and play an essential role in the defense against pathogens and in immune regulation. They develop from haematopoietic stem cells in the bone marrow and circulate in the blood and in various tissues, where they recognize and eliminate pathogenic microorganisms, foreign substances and abnormal cells. Through complex signaling mechanisms, leukocytes can specifically migrate into infected or inflamed tissue, trigger immune reactions and contribute to tissue regeneration. Their function and activity are regulated by a finely tuned network of cytokines and other immune modulators. Dysfunctions or quantitative deviations in the leukocyte count can be associated with immunological disorders, inflammatory processes or haematological diseases.
Leukocytes can be divided into three main groups:
- Lymphocytes (T-cells, B-cells, NK-cells) and
- Monoczytes,
which take on specific functions in the immune response.
The subpopulations of the individual leukocyte cell groups are extremely diverse and are subject to continuous change, as new protein structures, functions and cell interactions are constantly being discovered. In addition to histological classification, the cluster-of-differentiation (CD) nomenclature has long been used to characterize immune cells. Both the presence and absence of certain CD markers can be decisive for the differentiation of individual cell populations.
A common feature of all leukocytes is the expression of CD45 (CD45⁺). This surface protein, also known as receptor tyrosine protein phosphatase C, is exclusively expressed by all nucleated cells of hematopoiesis and is highly conserved.
Leukocytes or specific subgroups can be isolated and enriched using various methods.
