Use a pestle to grind sample material such as tissue or pieces of tissue on a pluriStrainer, pluriStrainer Maxi, Syringe Strainer, Re-Strainer or ÜbeStrainer. The sample material is comminuted by mechanical homogenisation between the pestle and the sieve mesh. This releases cells from a tissue, for example, while larger particles are retained.
Here is a simple guide: For homogenizing cells, tissues or food/feed samples:
Preparation:
- Select a suitable pluriStrainer (e.g. 70 µm or 100 µm pore size).
- Place the pluriStrainer on a suitable tube or in a Petri dish.
- If necessary, cool the tissue and tools to 4 °C to maintain cell viability.
Place the tissue on the pluriStrainer:
- Place the piece of tissue in the center of the pluriStrainer.
- If necessary, add a small amount of buffer solution (e.g. PBS or cell culture medium) to protect the cells.
Crush with the pestle:
- Use the pestle to gently grind the tissue with circular or back and forth movements.
- Apply gentle pressure to press the cells through the sieve without damaging them too much.
- If the tissue is too firm, additional buffer can be added to facilitate shredding.
Rinse & collect:
- Rinse the remaining material on the sieve with some buffer to catch all the cells that have passed through.
- Collect the filtered cell solution from the tube for further analysis or experiments.
Further processing:
- If necessary, centrifuge the cell suspension for further purification or to pellet the cells.
