pluriBead, cell separation without any magnetic components
pluriBead is a unique cell separation technology that works entirely without magnetic components. The process is simple: you sieve your pluriBeads (with bound target cells) through a sieve - the pluriBeads with your target cells stay on top, the unwanted cells pass through. After detaching, you have your target cells ready.
The flow-through is free of antibody-bound beads and can be used for further positive or negative cell enrichment. Enrichment with pluriBead can be performed without whole blood preparation. Contamination with unbound cells, especially platelets or erythrocytes, is very low if the cells are isolated directly from whole blood. The positive separation is faster, less time-consuming and leads to a higher purity of the target cells.
Worth knowing:
Positive cell separation vs. negative cell separation
By means of positive selection, a specific antibody binds directly to the desired cell type. The cells are "touched" by the antibody. All unwanted cells remain unbound and are separated from the labeled and desired cells in the subsequent enrichment steps. The easiest way to retain the labeled cells that are coupled to a solid phase is to use a cell strainer or magnets.
This contrasts with negative cell isolation. Here, the unwanted cells and cell types are marked, while the desired cells are "untouched". Magnetic activated cell separation (MACS) is often used for this purpose: Antibodies with magnetic particles bind to certain cells, which are then removed using a magnet.
Both techniques are used in immunology, stem cell research and cancer research to obtain pure cell populations for further analysis.
